Malacca / Melaka is my home state back home. I would say, depending on which part of KL you live in and how fast you drive, your journey there will take you about 1&1/2 to 2 hrs more or less.

There's a fair bit of things to see in Malacca. I'd say you can spend a day there which is enough to cover the hi-lights but staying overnight would help keep you rested and make it more enjoyable rather than to rush things along.

Then again, it depends on what kind of sights are you looking and what are things that attract you? Food? History? Cultural? Night scenes? etc etc.
